Preconditioning of Hypoxic Culture Increases The Therapeutic Potential of Adipose Derived Mesenchymal Stem Cells

چکیده

Various in vitro preconditioning strategies have been implemented to increase the regenerative capacity of MSCs. Among them are modulation culture atmosphere (hypoxia or anoxia), three-dimensional (3D), addition trophic factors (in form growth factors, cytokines hormones), lipopolysaccharides, and pharmacological agents. Preconditioning mesenchymal stem cells by culturing a hypoxic environment, which resembles natural oxygen environment tissues (1% –7%) not with standard conditions (21%), increases survival these via Hypoxia Inducible Factor-1α (HIF-1a) Akt-dependent mechanisms. In addition, precondition stimulates secretion pro-angiogenic expression chemokines SDF-1 (stromal cell-derived factor-1) its receptor CXCR4 (chemokine type 4) - CXCR7 7) engraftment cell. This review aims provide an overview preconditioned treatment therapeutic potential adipose-derived cells.

Evaluating the Viability of Adipose Tissue-derived Mesenchymal Stem Cells in Diabetic Culture Media Model

Introduction: In diabetes mellitus, stem cells are exposed to inappropriate conditions such as an increase of glucose in extracellular space, oxidative stress, and deprivation from growth factors (following ischemia). This study was aimed to evaluate the effect of these conditions on stem cells. Methods: Stem cells were isolated from subcutaneous adipose tissue of rats. The cells were maintaine...

Comparative analysis of the Gene expression profile of Chemokine Receptors between Adipose-derived and Bone marrow-derived Mesenchymal Stem Cells

Introduction: Mesenchymal stem cells (MSCs) hold great promise in the field of regenerative medicine.Although originally isolated from bone marrow, MSCs have since been obtained from a variety of adult and neonatal tissues including the adipose tissue. Stemness and multipotential features of Mesenchymal Stem Cells (MSC) has been highlighted in many studies but there are many dark aspects in ex...

Reprogramming by cytosolic extract of human embryonic stem cells improves dopaminergic differentiation potential of human adipose tissue-derived stem cells

The extract of pluripotent stem cells induces dedifferentiation of somatic cells with restricted plasticity. In this study, we used the extract of human embryonic stem cells (hESC) to dedifferentiate adipose tissue-derived stem cells (ADSCs) and examined the impact of this reprogramming event on dopaminergic differentiation of the cells. For this purpose, cytoplasmic extract of ESCs was prepare...

Enhanced neuro-therapeutic potential of Wharton's Jelly-derived mesenchymal stem cells in comparison with bone marrow mesenchymal stem cells culture.

Substantial inconsistencies in mesenchymal stem (stromal) cell (MSC) therapy reported in early translational and clinical studies may indicate need for selection of the proper cell population for any particular therapeutic purpose. In the present study we have examined stromal stem cells derived either from umbilical cord Wharton's Jelly (WJ-MSC) or bone marrow (BM-MSC) of adult, healthy donors...
